Moore moved to New York City in 1983, working as a waitress before being cast in the dual roles of Frannie Hughes and Sabrina Hughes on the soap opera As the World Turns, for which she won a Daytime Emmy Award; she played the roles from 1985 to 1988.

Moore began starring in feature films in the early 1990s, mostly appearing in supporting roles in films like The Hand That Rocks the Cradle, Benny and Joon, and The Fugitive. Her part in 1993's Short Cuts gained her critical acclaim and recognition, and she was cast in several high-profile Hollywood films, including 1995's romantic comedy Nine Months, and 1997's summer blockbuster The Lost World: Jurassic Park. Her role in the well-reviewed independent film Safe also attracted critical attention.

During the late 1990s and early 2000s, Moore appeared in a series of films that received Oscar recognition, including her roles in Boogie Nights (Best Supporting Actress nomination), The End of the Affair (Best Actress nomination) and her two 2002 films, Far From Heaven (Best Actress nomination) and The Hours (Best Supporting Actress nomination). During this period, she also appeared in the commercial successes Hannibal and The Forgotten and in follow-up to Boogie Nights, Magnolia.

Her film Freedomland opened in February 2006 to mixed reviews. Another film, Trust the Man, is directed by her husband and also features her son, Caleb. In March 2006, it was announced Moore would make her Broadway debut in the world premiere of David Hare's new play The Vertical Hour. The play opened in November 2006 and was directed by Sam Mendes. Also in 2006, Moore appeared in the film Children of Men. She most recently appeared in Next and the controversial incestuous film Savage Grace.

She played the girlfriend of her husband's character "Terl" in the 2000 film Battlefield Earth, and received "Worst Supporting Actress" at the 21st Golden Raspberry Awards for her role in the film.

In 2008 she was cast in a television pilot for a potential show called Suburban Shootout. She previously had a recurring role on Medium.

She debuted off-Broadway at Second Stage Theatre in the role of 'Holly Dancer' in Tina Howe's widely acclaimed Coastal Disturbances (1987). However, despite the praise and recognition, it took some time for that success to translate to her film career. Her television debut was with the made-for-TV movie Manhunt for Claude Dallas (1986). Her first major role in a theatrical feature was in The Great Outdoors (1988) playing 'Kate Craig'. Her next role was as the Marquise de Merteuil in Valmont (1989) opposite Colin Firth.

Bening's next major feature, Stephen Frears's The Grifters (1990) met with critical acclaim. She received an Academy Award nomination for Best Supporting Actress for The Grifters. She followed that with her appearance in Bugsy. Bening was paid $3 million to play the role of 'Elise Kraft/Sharon Bridger' in The Siege (1998). Her next role, in the 1999 film American Beauty, would give her the highest-profile role of her career thus far. Other movies: Regarding Henry, What Planet Are You From?, Postcards From The Edge...

She replaced Julianne Moore in her most recent appearance, Running with Scissors, playing Deirdre Burroughs in the film adaptation of the Augusten Burroughs book.

De Mornay's film debut was a small part in One from the Heart (1982). Soon thereafter came her star-making role as a hooker who seduces a high school student played by Tom Cruise in Risky Business. De Mornay's most commercially successful film since then came in the thriller The Hand That Rocks the Cradle (1992). She also appeared in Ron Howard's Backdraft, in a remake of Roger Vadim's provocative And God Created Woman and in the starring role as a defense lawyer in murder drama Guilty as Sin.

In 2004, the actress guest-starred as attorney Hannah Rose for the final few episodes of The Practice and the following year had a brief role in Wedding Crashers. In June 2007, she appeared in the HBO series John From.
