Foster made nearly 50 film and television appearances before she attended college. She debuted as a television actress in a 1968 episode of Mayberry R.F.D. In 1969, she appeared in an episode of Gunsmoke. Although not a regular on The Courtship of Eddie's Father, she appeared from time to time. She made her film debut in the 1970 TV movie Menace on the Mountain. Foster made a number of Disney movies, including Napoleon and Samantha (1972), One Little Indian (1973), Freaky Friday (1976), and Candleshoe (1977). She also co-starred with Christopher Connelly in the 1974 TV series version of Paper Moon and in the 1976 cult film The Little Girl Who Lives Down the Lane.

At age 14, Foster was nominated for the Academy Award For Best Supporting Actress for her role in Taxi Driver. Foster received two BAFTA awards in 1976: Best Newcomer and Best Supporting Actress for her performances in Bugsy Malone and Taxi Driver. Foster successfully made the transition to adult roles, but not without initial difficulty. Several of her post-Taxi Driver works were financially unsuccessful, such as Foxes, The Hotel New Hampshire, Five Corners, and Stealing Home. She had to audition for her role in The Accused. She won the part and the first of her two Golden Globes and Academy Awards as Best Actress for her role as a rape survivor. She earned her second as FBI agent Clarice Starling in the 1991 film The Silence of the Lambs.

Foster played in Sommersby and in1994 film Maverick. In 1997, she starred in the sci-fi movie Contact. In 2002, Foster got lead role in Panic Room. The film grossed over 30 million dollars in its opening weekend in the United States, Foster's biggest box office opening success of her career so far. She then performed in the French-language film Un long dimanche de fiançailles (A Very Long Engagement) (2004), speaking French fluently throughout. Foster returned in the 2005 film Flightplan which opened once again #1 at the U.S. box office and was a worldwide hit.

In 2006, she appeared in Inside Man which opened #1 at the U.S. box office and went on to be one of the commercial hits of the year. In 2008, she starred in The Brave One, another urban thriller that opened at #1 at the U.S. box office. Foster's performance in the film would earn her a sixth Golden Globe acting nomination.

In 2008, Foster starred in Nim's Island, the first comedy that Foster has starred in since Maverick in 1994. Foster is currently developing a biopic of Leni Riefenstahl.

Moore moved to New York City in 1983, working as a waitress before being cast in the dual roles of Frannie Hughes and Sabrina Hughes on the soap opera As the World Turns, for which she won a Daytime Emmy Award; she played the roles from 1985 to 1988.

Moore began starring in feature films in the early 1990s, mostly appearing in supporting roles in films like The Hand That Rocks the Cradle, Benny and Joon, and The Fugitive. Her part in 1993's Short Cuts gained her critical acclaim and recognition, and she was cast in several high-profile Hollywood films, including 1995's romantic comedy Nine Months, and 1997's summer blockbuster The Lost World: Jurassic Park. Her role in the well-reviewed independent film Safe also attracted critical attention.

During the late 1990s and early 2000s, Moore appeared in a series of films that received Oscar recognition, including her roles in Boogie Nights (Best Supporting Actress nomination), The End of the Affair (Best Actress nomination) and her two 2002 films, Far From Heaven (Best Actress nomination) and The Hours (Best Supporting Actress nomination). During this period, she also appeared in the commercial successes Hannibal and The Forgotten and in follow-up to Boogie Nights, Magnolia.

Her film Freedomland opened in February 2006 to mixed reviews. Another film, Trust the Man, is directed by her husband and also features her son, Caleb. In March 2006, it was announced Moore would make her Broadway debut in the world premiere of David Hare's new play The Vertical Hour. The play opened in November 2006 and was directed by Sam Mendes. Also in 2006, Moore appeared in the film Children of Men. She most recently appeared in Next and the controversial incestuous film Savage Grace.
