"Despeluque" comes from the verb "despelucar", which literally means "to mess one's hair". But in the context of the sentence, and just like "irse de rumba" which I translated to "to party", "ir de despeluque" is another slang for "to party". But those are Colombian slangs, so forgive me if I'm wrong. I had to do a quick research on this. We don't even use the word "despelucar" in Mexico to describe messing somebody's hair, we call that "despeinar".
